The Spanish police said that a vehicle was off a highway and burst into flames in the northern province of Zamora shortly after midnight.
Jota, 28, married his partner Rute Cardoso on 22 June. The couple had three children.
He spent the last five years in Anfield and helped the English giants win the Premier League last season.
“Liverpool Football Club was destroyed by the tragic death of Diogo Jota,” the club said in a statement.
“The club has been informed that the 28-year-old died after a traffic accident in Spain together with his brother, Andre.
“Liverpool FC will currently not give any further comments and the privacy of Diogo and Andre's family, friends, teammates and club staff are asked, will be respected if they try to deal with an unimaginable loss.
“We will continue to offer them our full support.”
